Study on the effect of dissipation and discharge method on intestinal microflora in bronchial asthmatic rats based on 16S rDNA gene sequencing technology
Objective To study the effect of Baiguo Wendan Decoction on the intestinal flora structure of rats with bronchial asthma by the mothod of the 16S ribosomal DNA gene sequencing technology.Methods Seventy-two healthy male SD rats were randomly divided into blank group,model group,dexamethasone group,high,medium,and low dose groups of Baiguo Wendan Decoction,with 12 rats in each group.Preparation of asthma rat model by sensitization and challenge with OVA.During the experiment,observe and record the changes in the mental state,food consumption,water consumption,respiration,and hair gloss of rats in each group.Observed the pathological changes of HE staining in lung tissue of rats in each group.The method of 16S rDNA Miseq high throughput sequencing was used to view intestinal flora.Analyzed the diversity of intestinal flora through OTU cluster analysis,Alpha diversity analysis,and Beta diversity analysis methods.Comparison of differences in intestinal microflora structure of rats under different intervention methods by SPSS 25.0 analysis.Results(1)Behavioral observation results:the mental state,diet,water consumption,hair,and respiration of rats in the high,medium,and low dose groups of Baiguo Wendan Decoction and the dexamethasone group were improved to varying degrees.(2)In the model group,the pulmonary tracheal epithelium was damaged,the pulmonary septum thickened,and inflammatory cells were increased.Compared with the model group,each treatment group had some improvement.(3)After modeled,compared with the blank group,the model group's intestinal flora abundance,Chao1 index,and Observed_species index,Faith's PD index,Pielou_e index and Shannon index have a downward trend.In the dexamethasone group,each index of intestinal flora in rats were decreased significantly,with a statistically significant difference(P<0.05).Compared with the dexa-methasone group,the low and medium dose groups of Baiguo Wendan Decoction,Pielou_e index and Shannon index were increased significantly,with a statistically significant difference(P<0.05).The Chao1 index,Observed_species index,Faith's PD index and Shannon index of Baiguo Wendan Decoction high dose group was increased significantly,and the difference was statistically significant(P<0.05).Compared with the low dose group of Baiguo Wendan Decoction,the Faith's PD index in the high dose group was significantly increased with a statistically significant difference(P<0.05).(4)After beta diversity analysis,there were significant differences in intestinal flora between the blank group and the model group,between the blank group and the dexamethasone group,between the model group and each treatment group,between the dexamethasone group and each dose group of Baiguo Wendan Decoction,between the low dose group of Baiguo Wendan Decoction and the medium and the high dose groups of Baiguo Wendan Decoction(P<0.05).(5)At the family level,the relative proportion of Lactobacillaceae and Clostridiaceae in the model group were increased,while the relative proportion of Desulfovibrionaceae,Digestive Streptococcaceae,Ruminococcaceae,and S24-7 were decreased.Conclusion Baiguo Wendan Decoction can improve the inflammatory infiltration of lung tissue in asthmatic rats,and its mechanism may be related to its restoration of intestinal flora diversity,richness and regulation of flora structure.
