清热化湿调枢方对葡聚糖硫酸钠结肠炎模型小鼠肠黏膜MUC-2、Claudin-2表达的影响

目的 本研究基于粘蛋白2(mucoprotein-2,MUC-2)和闭合蛋白2(Claudin-2)的表达变化,探讨了在预防和缓解小鼠肠黏膜损伤的DSS结肠炎模型中,清热化湿调枢方可能发生的机理.方法 24只C57BL/6小鼠随机抽取6只小鼠为正常组,其余18只小鼠均给予2.5%葡聚糖硫酸钠(dextran sodium sulfate,DSS)水溶液自由饮用7天以制成每组6只的DSS结肠炎小鼠模型,然后再将其随机分为模型组,美沙拉嗪组,清热化湿调枢方组.对正常组、模型组的小鼠,每组用0.5 mL的纯净水进行灌胃;美沙拉嗪组的小鼠给美沙拉嗪混悬液灌胃,剂量为0.52 g/(kg·d);清热化湿调枢方组的小鼠给予1.69 g/(kg·d)的清热化湿调枢方中药颗粒剂灌胃;每组每天1次,共7天,与模型同步进行.7天后测定各组小鼠体质量,记录小鼠体质量变化,评分疾病活动指数(disease activity index,DAI),记录各组小鼠结肠长度,观察结肠组织病理变化,检测MUC-2和Claudin-2在结肠组织中的表达.结果 模型组的小鼠体质量较正常组显著下降,DAI评分显著上升,结肠长度变短,结肠组织中Claudin-2和MUC-2蛋白的表达明显下降(P<0.05).与模型组相比,美沙拉嗪组和清热化湿调枢方组的小鼠体质量显著增加,DAI评分下降,结肠长度增加,Claudin-2、MUC-2蛋白在结肠组织中的表达均有明显提高(P<0.05).从结肠组织的病理变化来看,模型组小鼠肠黏膜上皮可见明显缺损,杯状细胞减少,固有层和隐窝结构破坏严重,排列错乱,淋巴细胞浸润较多;清热化湿调枢方组和美沙拉嗪组小鼠结肠黏膜上皮近乎完整,炎性细胞少量浸润.结论 清热化湿调枢方对DSS结肠炎小鼠肠道黏膜屏障损伤具有一定防治作用,其机制可能是通过提高结肠组织MUC-2表达,刺激Claudin-2分泌促进黏膜修复,从而维持肠黏膜屏障的稳态.
Effect of Qingre Huashi Tiaoshu Recipe on the expression of MUC-2 and Claudin-2 in intestinal mucosa of DSS colitis model mice
Objective Based on the changes of mucoprotein-2(MUC-2)and Claudin-2 expression,this study explored the possible mechanism of Qingre Huashi Tiaoshu Recipe in preventing and alleviating intestinal mucosal barrier damage in dextran sodium sulfate(DSS)colitis model mice.Methods 24 C57BL/6 mice were randomly selected as the normal group,and the remaining mice were given 2.5%DSS aqueous solution to drink freely for 7 days to make molds,and then randomly divided into model group,mesalazine group and Qingre Huashi Tiaoshu Recipe group,with 6 mice in each group.Mice in the normal group and the model group were given 0.5 mL of pure water for gastric gavage in each group;mice in the mesalazine group were given 0.52 g/(kg·d)mesalazine suspension for gavage;mice in the Qingre Huashi Tiaoshu Recipe group were given 1.69 g/(kg·d)gavage.Gastric gavage was performed once a day for 7 days in each group at the same time as molding.After 7 days,the body weight changes of each group were measured and recorded,the disease activity index(DAI)of each group was scored,the colon length of each group was measured,pathological changes of colon tissue were observed,and the ex-pressions of MUC-2 and Claudin-2 in colon tissue were detected.Results Compared with the normal group,body weight,DAI score,colon length and Claudin-2 and MUC-2 protein expressions in colon tissue were significantly decreased in the model group(P<0.05).Compared with the model group,body weight,DAI score,colon length and MUC-2 protein expression were significantly increased in mesalazine group and the Qingre Huashi Tiaoshu Recipe group(P<0.05).In terms of the pathological changes of colon tissue,the intestinal mucosal epithelium of mice in the model group was defective,the goblet cells were significantly decreased,the structure of lamina propria and crypt was seriously damaged,the arrangement was disordered,and a large number of lymphocytes infiltrated.There was almost no defect in colonic mucosal epithelium of mice in the heat-clearing and moist-clearing the Qingre Huashi Tiaoshu Recipe group and the mesalazine group,and a few inflammatory cells infiltrated.Conclusion Qingre Huashi Tiaoshu Recipe has certain preventive and palliative effects on intestinal mucosal barrier damage in DSS colitis mice,which may be caused by increasing the expression of MUC-2 in colon tissue,stimulating Claudin-2 secretion and promoting mucosal repair,so as to maintain the homeostasis of intestinal mucosal barrier.
