Exploring the effect of Sanjiao acupuncture on diarrheal irritable bowel syndrome rats based on SCF/C-kit pathway
Objective To observe the effects of Sanjiao acupuncture on the abnormal gastrointestinal dynamics and the expression of SCF and C-kit mRNA and protein in colon tissue of Irritable bowel syndrome with diarrhea(IBS-D)rats,and to explore the mechanism of action of Sanjiao acupuncture in the treatment of IBS-D.Methods Wistar male rats were randomly divided into blank group,model group,Western medicine group and Sanjiao group,6 rats in each group.Except for the blank group,all the other groups were treated with"glacial acetic acid enema+restraint stress"to establish the IBS-D rat model.The rats were treated for 14 days.The general status of the rats was observed;body mass,open field test(lateral and longitudinal movements of rats),faeces,AWR3 time-lapse water injection,Real Time-PCR,Western blot,and the number of rats were measured.The method of PCR,Western blot were used to detect the levels of SCF,C-kit mRNA and protein expression in colonic tissue.Results Compared with the blank group,the general condition of IBS-D rats was poorer,body mass was significantly lower(P<0.01),the distance of lateral movement and the number of longitudinal movements were significantly lower(P<0.01),rate of faeces was significantly higher(P<0.01),AWR3 water injection was significantly lower(P<0.01)and SCF,C-kit mRNA and protein expression levels in colonic tissues were significantly higher(P<0.01),and protein expression levels were significantly increased in colon tissue(P<0.01).Compared with the model group,the body mass of the rats in the Western medicine group and the Sanjiao group was significantly higher(P<0.01,P<0.05),the distance of lateral movement and the number of lon-gitudinal movements were significantly higher(P<0.05,P<0.01),the rate of faeces was significantly down-regulated(P<0.01),the AWR3 time-lapse water injection was significantly higher(P<0.01),and the expression of SCF,C-kit and protein levels in the colon tissue of the rats in the Sanjiao group were significantly higher(P<0.01),mRNA and protein level expression were significantly decreased(P<0.01),while SCF,C-kit mRNA and protein expression in the Western medicine group were not statistically significant(P>0.05).Conclusion Sanjiao needle method can improve the symptoms of IBS-D rats,which may be related to the regulation of SCF,C-kit mRNA and protein expression levels in colon tissue.
