川芎提取液制备工艺的优化及免疫调节作用研究

目的 优化川芎提取液的制备工艺,研究川芎提取液对小鼠的安全性和对免疫抑制小鼠的免疫活性.方法 首先,采用L9(34)正交试验法,以阿魏酸和川芎多糖的转移率为考察指标,对提取时间、提取次数、加水量等3个因素进行考察,优化川芎的提取工艺.进一步,将川芎提取液浓缩并分成两部分,一部分浓缩,一部分浓缩后醇沉,合并浓缩液与醇沉物,以外观性状和口感为指标,比较确定川芎浓缩液和醇沉液的比例,制备川芎提取液.随后,采用腹腔注射环磷酰胺制备小鼠免疫抑制模型,灌胃给予小鼠川芎提取液低、中、高剂量,其浓度分别为190.0、380.0、570.0 mg/kg,连续灌胃32天后取材,测定小鼠脏器指数;酶联免疫吸附法(enzyme-linked immunosorbent assay,ELISA)检测小 鼠免疫球蛋白 G(immunoglobin G,IgG)、免疫球蛋白 M(immunoglobin M,IgM)、补体 C3(complement 3,C3)、补体 C4(complement 4,C4)、白细胞介素-6(interleukin-6,IL-6)、白细胞介素-8(interleukin-8,IL-8)、肿瘤坏死因子 α(tumor necrosis factor-α,TNF-α)水平的变化;MTT法检测脾淋巴细胞的转化率;LDH法测自然杀伤细胞(natural killer cell,NK)活性.结果 阿魏酸和川芎多糖的最佳提取工艺参数为提取2次,第一次提取液料比为10∶1,提取时间为4小时;第二次提取液料比为5∶1,提取时间为2小时.川芎水提浓缩液与65%水提醇沉物配比为3∶2时,制备的川芎提取液溶液澄清度高、口感好.与模型组比较,胸腺指数和脾脏指数均显著上升(P<0.01,P<0.001),川芎提取液各剂量组IgG、IgM、C3、C4含量均显著升高(P<0.001,P<0.01,P<0.05),川芎高剂量组IL-6、IL-8、TNF-α含量均显著降低(P<0.001,P<0.01,P<0.05),川芎中剂量组IL-6、IL-8含量均显著降低(P<0.001),川芎低剂量组IL-8含量显著降低(P<0.01),川芎提取液各剂量组均可显著提高免疫低下动物NK细胞活性和脾淋巴细胞转化率(P<0.001,P<0.01,P<0.05).结论 优化的川芎提取液制备工艺可行、简便、稳定、澄清度高、口感好、安全性高,对环磷酰胺致免疫抑制小鼠具有免疫增强作用.
Optimization of the preparation process of Chuanxiong extract and study on its immunomodulatory effect
Objective Optimize the preparation process of Chuanxiong extract and to study the safety and immune activity of Chuanxiong extract on immunosuppressed mice.Methods Firstly,the L9(34)orthogonal experimental method was used to optimize the extraction process of Ligusticum Chuanxiong by examining three factors:extraction time,extraction frequency,and water addition,with the transfer rate of ferulic acid and Ligusticum Chuanxiong polysaccharides as the evaluation indicators.Furthermore,the extract of Ligusticum Chuanxiong was concentrated and divided into two parts.One part was concentrated,the other part was concentrated and then alcohol precipitated.Merge concentrated solution and alcohol precipitate,and the ratio of Ligusticum Chuanxiong concentrate and alcohol precipitate was compared and determined based on appearance and taste as indicators to prepare the extract of Ligusticum Chuanxiong.Subsequently,the mice immunosuppressive model was prepared by intraperitoneal injection of cyclophosphamide.The low,medium,and high doses of Chuanxiong extract in mice were administered by gavage at 190.0,380.0 and 570.0 mg/kg,respectively.After continuous gavage for 32 days,the mouse organ index was measured,and the ELISA method for detecting mouse immunoglobulins(IgG,IgM),complement C3,C4,interleukin-6(IL-6),interleukin-8(IL-8),and tumor necrosis factor(TNF-α)Horizontal changes,MTT method to detect the transformation rate of splenic lymphocytes,and LDH method to measure NK cell activity.Results The optimal extraction process parameters for ferulic acid and Chuanxiong polysaccharides are twice,with a liquid to material ratio of 10∶1 for the first extraction and an extraction time of 4 hours;the second extraction had a liquid to material ratio of 5∶1 and an extraction time of 2 hours.When the ratio of Chuanxiong water extract concentrate to the lower layer of 65%water extract and alcohol precipitation is 3∶2,the prepared Chuanxiong extract solution has high clarity and good taste.Compared with the model group,the thymus index and spleen index were significantly increased(P<0.01,P<0.001),and the IgG,IgM,C3,and C4 contents in each dose group of Chuanxiong extract were significantly increased(P<0.001,P<0.01,P<0.05).The high-dose group of Chuanxiong extract had IL-6,IL-8,and TNF-α levels.The content was significantly reduced(P<0.001,P<0.01,P<0.05),The content of IL-6 and IL-8 in the medium dose group of Chuanxiong was significantly reduced(P<0.001),while the content in the low dose group of Chuanxiong was significantly reduced(P<0.01),the levels of NK cell activity and spleen lymphocyte transformation rate in immunocompromised animals in all the three dose groups of Chuanxiong extract were significantly increased(P<0.001,P<0.01,P<0.05).Conclusion The optimized preparation process of Chuanxiong extract is feasible,simple,stable,with high clarity,good taste and high security;It has an immunomodulatory effect on immunosuppressive mice induced by cyclophosphamide.
