The antitumor effect of Sinocrassulosides Ⅵ/Ⅶ and its effect on apoptosis and cell cycle of BGC-823 cells
Objective To study the effect of Sinocrassulosides Ⅵ/Ⅶ on the proliferation of HepG-2、BGC-823、HT-29 tumor cell lines,and to detect its effect on apoptosis and cell cycle of BGC-823 gastric cancer cell line.Methods HepG-2,BGC-823 and HT-29 cells were treated with different concentrations of Sinocrasulosides Ⅵ/Ⅶ(80,40,20,10,5 μg/mL)for 24,48,72 h,and then cell proliferation was detected by MTT assay.The cell cycle and apoptosis rate of BGC-823 cell line were detected by flow cytometry.Results The MTT results showed that Sinocrasulosides Ⅵ/Ⅶ could significantly inhibit the proliferation of HepG-2,BGC-823 and HT-29 cells in a concentration-dependent manner compared with the control group,and BGC-823 cell line was the most effective against Sinocrasulosides Ⅵ/Ⅶ.Flow cytometry showed that Sinocrasulosides Ⅵ/Ⅶ could arrest BGC-823 cells in S phase,G2/M phase,inhibit G0/G1 phase and promote apoptosis of BGC-823 cells from 1.5%to 8.6%(P<0.01).Conclusion Sinocrasulosides Ⅵ/Ⅶ can inhibit the proliferation of HepG-2,BGC-823,HT-29 cells,and the mechanism of inducing apoptosis of BGC-823 cells may be related to cell cycle arrest.
