Mass spectrometry analysis combined with network pharmacology and animal experiments to investigate the mechanism of action of Jiannao Guben Formula in the treatment of cerebral small vessel disease cognitive impairment
Objective To investigate the mechanism of action of Jiannao Guben Formula for the treatment of cerebral small vessel disease cognitive impairment(CSVCI)with the method of mass spectrometry,network pharmacology and animal experiments.Methods The main active ingredients were identified by mass spectrometry,and their corresponding targets were searched by TCMSP database;the targets of CSVCI were searched by GeneCards and OMIM databases;the network of drug targets was visualised by String database and Cytoscape software;the intersecting targets were analyzed by GO and KEGG using DAVID database.Results The active ingredients,including kaempferol and diosgenin,acted on the core targets of MAPK8 and AKT1,and may be involved in the inflammatory response through the PI3K-Akt signalling pathway,etc.The results of WB showed that Jiannao Guben Formula inhibited the expression of Notch-1,RBP-Jκ and Hes-1.Conclusion Jiannao Guben Formula can pass through multiple targets and pathways to treat CSVCI,and it may inhibit the inflammatory response through the Notch-1 signaling pathway for the treatment of CSVCI.
