Mechanism of deer penis and tortoise shellmedicine in nourishing yin and tonifying kidney to relieve physical fatigue
Objective This study aimed to investigate the effect and mechanism of deer penis and tortoise shell on nourishing yin and tonifying kidney to alleviate physical fatigue in mice with kidney yin de-ficiency.Methods After adaptive feeding for one week,70 healthy male Kunming mice were randomly divided into 7 groups,including the blank group,the kidney yin deficiency model group,the Zuoguiwan medicine group,the low-dose group of deer penis-tortoise shell medicine,the high-dose group of deer penis-tortoise shell medicine,the deer penis group,and the tortoise shell group,with 10 mice in each group.The modeling and administration were conducted simultaneously for a total of 14 days.The exhaustion swimming time was measured 30 minutes after drug administration on the 14th day;On the 15th day,the eyeball was removed and blood was collected to separate serum and measure the levels of lactate,blood urea nitrogen,testosterone,Cortisol,cyclic adenosine monophosphate(cAMP),and cyclic guanosine monophosphate(cGMP).The liver was dissected to measure the content of liver glycogen and the protein expression of phosphoinositide 3 kinase(PI3 K),protein kinase B(Akt),and glycogen synthase kinase-3(GSK-3 β).Results Compared with the model group,the body weight of the deer penis-tortoise shell drug group and the monotherapy group significantly was increased(P<0.01),the exhaustion swimming time was increased significantly(P<0.05,P<0.01),the blood lactate content was decreased significantly(P<0.01),the blood urea nitrogen content significantly was decreased(P<0.01),the liver glycogen content was increased significantly(P<0.05,P<0.01),the testosterone content was increased significantly(P<0.01),and the cGMP content was increased significantly(P<0.01),cAMP/cGMP ratio was significantly decreased(P<0.01),liver Akt protein expression was significantly increased(P<0.05),and liver GSK-3 β protein expression was significantly decreased(P<0.05),The expression level of PI3K protein in high dose group and deer whip group was significantly increased(P<0.05).Conclusion Deer penis and tortoise shell medicine can increase glycogen synthesis by affecting PI3K/Akt/GSK-3β pathway,thereby nourishing yin and tonifying the kidneys and relieving physical fatigue.
