Effect of Bushen Jieyu Tiaochong Decoction on reproduction,metabolism and stress in chronic stress PCOS model rat
Objective The present study aims to investigate the therapeutic effects of Bushen Jieyu Tiaochong Decoction on reproductive endocrine function,glucose metabolism,and chronic stress in a rat model induced by letrozole combined with chronic unpredictable mild stimulation(CUMS).Methods Thirty-five female SD rats were selected for the experiment,and 30 rats with a normal estrous cycle were randomly divided into six groups:blank group,model group,western medicine group,low-dose traditional Chinese medicine(TCM)group,medium-dose TCM group,and high-dose TCM group.Each group consisted of 5 rats.Except for the blank group,all rats were administered letrozole combined with CUMS for 42 days to establish the model.After 42 days of modeling,rats in the low-dose,medium-dose and high-dose TCM groups were received different doses of Bushen Jieyu Tiaochong Decoction while rats in the western medicine group was given Diane-35 combined with fluoxetine hydrochloride dispersible tablets.After 28 days of treatment,behavioral tests and insulin tolerance test were conducted.The serum levels of luteinizing hormone(LH),follicle stimulating hormone(FSH),free testosterone,and insulin were measured using enzyme-linked immunosorbent assay.The method of Hematoxylin and eosin staining was used to observe pathological and morphological changes in ovarian tissue.High-performance liquid chroma-tography-electrochemical detection was employed to detect the expression of monoamine neurotransmitters in hippocampus.Results Compared with the control group,the model group exhibited significant increases in ovarian volume,weight,and index(P<0.05),along with polycystic changes and a significantly elevated LH/FSH ratio(P<0.05).In terms of metabolism,the area under the curve of the insulin tolerance test was increased significantly(P<0.05),as well as fasting blood glucose levels and HOMA-IR(P<0.05).The levels of chronic stress,sucrose preference were decreased(P<0.05),open field test activity was decreased(P<0.05),and there was a tendency for neurotransmitter levels in hippocampal tissues to decrease(P>0.05).The western medicine group and various TCM groups showed varying degrees of improvement compared with the model group in terms of ovarian pathology,glucose metabolism,and chronic stress state.Conclusion The Bushen Jieyu Tiaochong Decoction can regulate monoamine neurotransmitters in the brain to reduce chronic stress state and improve reproductive function in PCOS patients while also regulating abnormal glucose metabolism associated with PCOS.
