Hegantang Granules improve duodenal low-grade inflammation of functional dyspepsia by regulating NF-κB signaling pathway
Objective To explore the mechanism of Hegantang in improving duodenal low-grade inflammation in functional dyspepsia(FD).Methods 18 6-week-old rats were randomly divided into the normal control,FD and Hegantang group,with 6 rats in each group.The rats in the FD group and the Hegantang group were given tail-clip stimulation method.The Hegantang group was given clinical equivalent dose of Hegantang granules by gavage.The stomach and duodenum of the rats were taken for hematoxylin-eosin staining,toluidine blue staining and eosinophilic staining to detect the tissue morphology and the aggregation of inflammatory cells.The expression of inflammatory factors,NF-κB P65 and p-P65 were detected by the methods of qPCR,ELISA,Western blot and immunofluorescence.Results Compared with the normal control group,the aggregation of mast cells and eosinophils in the FD group was more significant(P<0.01),the expression of pro-inflammatory factor and anti-inflammatory factor was increased(P<0.01),and the expression level of NF-κB P65 and p-P65 in duodenum were up-regulated(P<0.01).After Hegantang treatment,the aggregation of inflammatory cells and the expression of inflammatory factors were decreased(P<0.01),and the content of P65,p-P65 were down-regulated(P<0.05,P<0.01).Conclusion In the condition of FD duodenal microinflammation,NF-κB signaling pathway is partially activated along with the imbalance of pro-inflammation and anti-inflammation.Hegantang can down-regulate the expression and activation of NF-κB P65 and restore the balance of anti-in-flammatory and pro-inflammatory factors,which may be part of the molecular biological mechanism of Hegantang to improve FD duodenal low-grade inflammation.
