Effect of Jianpi Zhidong Decoction on the expression of histamine receptors HRH1,HRH2 and HRH3 in the striatum of TS rats
Objective To explore the effect of Jianpi Zhidong Decoction(JPZDD)on the expression of histamine receptors HRH1,HRH2,and HRH3 in the striatum of tourette syndrome(TS)model rats.Methods 24 SD rats were randomly divided into the blank group(6 rats)and the model group,with 18 rats in each group.The model group was injected with IDPN to establish the TS model.According to behavioral scores and the total distance of autonomous activity,they were randomly divided into the model group,the JPZDD group and the western medicine group,with intervention of 6 weeks.After the intervention,behavioral evaluations(stereotyped behavior and autonomous activity)were conducted,and the mRNA and protein expressions of HRH1,HRH2 and HRH3 in the striatum were detected using the methods of RT-PCR,Western blot,and immunohistochemistry.Results(1)After intervention,JPZDD significantly reduced the behavioral scores(P<0.01)and total distance of autonomous activity(P<0.05)when compared with the model group;(2)After intervention,there was no statistically significant difference in the protein and mRNA expressions of HRH1 and HRH2 in the JPZDD and Western medicine groups when compared with the model group;Compared with the model group,JPZDD significantly reduced the protein expression of HRH3(P<0.05 in Western blot and P<0.01 in IOD value)and mRNA expression of HRH3 in the striatum(P<0.05).Conclusion Jianpi Zhidong Decoction exerts anti tic effects by regulating the expression of HRH3 in the striatum of TS rats.
