绿萼梅水提物的工艺优化及其抗氧化研究

文章以绿萼梅为原料、多糖提取率为响应值,通过单因素试验并结合响应面法优化绿萼梅水提物的提取工艺,测定了不同产地、不同花期绿萼梅的多糖含量,并进行体外试验评价了不同产地、不同花期绿萼梅的抗氧化活性.结果表明,优化后的绿萼梅水提物的提取工艺为:提取时间1h、提取温度80℃、提取次数为3次、料液比1∶20(g/mL),该提取条件下绿萼梅多糖含量为11.13%,且多糖含量与其产地、花期密切相关.其中花期的影响更显著,盛开期的多糖含量最高,初开期次之,花蕾期最低.体外抗氧化活性实验表明,绿萼梅水提物具有较强的抗氧化活性,且产地、花期均影响其抗氧化活性.总体上花蕾期的抗氧化活性最强,明显强于盛开期;而不同产地之间则无明显规律.
Study on Optimization of Extraction Process and Antioxidation of Water extracts from Prunus Mume
In the article,the extraction process of water extract from Prunus mume was optimized by single factor test and response surface method with Prunus mume as raw material and polysaccha-ride extraction rate as response value.The polysaccharide content of Prunus mume in different produc-ing areas and different flowering periods were determined,and the antioxidant activity of Prunus mume in different producing areas and different flowering periods were evaluated in vitro.The results showed that the optimized extraction process of the water extracts of Prunus mume was as follow:extraction time of 1 h,extraction temperature of 80 ℃,extraction number of 3 times,material-liquid rate of 1:20(g/mL).The content of polysaccharide from Prunus mume was 11.13%under this condition,and the content of polysaccharide was closely related to its area and flowering stage.The polysaccha-ride content in the blooming stage was highest,followed by the initial opening period,and the lowest in the bud period.The in vitro antioxidant activity showed that the water extracts of Prunus mume had strong antioxidant activity,and the origin and flowering stage both influenced its antioxidant activity.In general,the antioxidant activity of the flower bud stage was the strongest and significantly stronger than that of the blooming stage;there was no significant pattern among different areas.
