Isolation of total RNA from mango pulp tissue rich in polysaccharides and polyphenols
The fruit of Zihua mango (Mangifera indica L.) was used as experimental material to improve the method for isolation of total RNA from mango pulp. This method, based on the modified Tirs-H_3BO_4 method, was improved by adding polyvinylpyrrolidone into the extraction solution, and the pre-cooling LiCl, isopropanol and ethanol absolute into the precipitation buffer. The results showed that total RNA precipitated by isopropanol was better than the other precipitants. Total RNA isolated by this method was of high quality with high ratio of yield and a full strip, and was able to be used for later molecular manipulations such as RT-PCR, Northen hybrid and cDNA library construction.
