金龟子绿僵菌制剂对二化螟的防效及体内相关酶活性的影响

用剂量为 1.6×107/mL 的金龟子绿僵菌 CQMa421 制剂处理二化螟幼虫,测定该制剂对二化螟幼虫的毒力及其对幼虫总蛋白含量及过氧化氢酶(CAT)、过氧化物酶(POD)、超氧化物歧化酶(SOD)等 3种保护酶活性和羧酸酯酶(CarE)、乙酰胆碱酯酶(AChE)、碱性蛋白酶(AKP)和谷胱甘肽 S-转移酶(GSTs)等 4 种解毒酶活性的影响.结果表明:经金龟子绿僵菌制剂处理后,二化螟幼虫的存活率呈持续下降趋势,半致死时间(LT50)为 7.23 d,第 10天死亡率为 56.89%,说明金龟子绿僵菌制剂对二化螟幼虫具有显著毒力;金龟子绿僵菌制剂处理 3 d 时,二化螟幼虫蛋白质含量最高;POD、CAT、SOD 等 3 种保护酶活性分别在处理 2、4、5 d 时达到最高,分别为198.76、19.59和 12.90 U/mg,均显著高于对照组相应酶的活性;4种解毒酶GSTs、AChE、CarE和AKP最高活性分别约为对照组的 1.68、1.68、1.27和 1.62倍,说明经金龟子绿僵菌制剂CQMa421处理后,二化螟幼虫体内稳态遭到破坏,进而受到毒害.
Effects of Metarhizium anisopliae preparation against Chilo suppressalis and on related enzyme activities in vivo
Chilo suppressalis(C.suppressalis)larvae were treated with Metarhizium anisopliae(M.anisopliae)CQMa421 preparation at the recommended field dose of 1.6×107 cells/mL.The toxicity of M.anisopliae CQMa421 preparation to C.suppressalis larvae as well as the larval total protein content and activities of catalase(CAT),peroxidase(POD),superoxide dismutase(SOD),carboxylesterase(CarE),acetylcholinesterase(AChE),alkaline protease(AKP)and glutathione S-transferases(GSTs)were determined.The results showed that the survival rate of C.suppressalis larvae showed a continuous decreasing trend after treatment with M.anisopliae CQMa421 preparation,preparation with the LT50 of 7.23 d and the mortality rate of 56.89%on the 10th day of treatment,indicating that the M.anisopliae CQMa421 preparation had significant toxicity to C.suppressalis larvae.The protein content of C.suppressalis larvae was the highest at 3 d of M.anisopliae CQMa421 preparation treatment.At 2,4 and 5 d after treatment,the activities of three protective enzymes,POD,CAT and SOD,reached the highest with 198.76,19.59 and 12.90 U/mg,respectively,which were significantly higher compared to the control.The highest activities of four detoxification enzymes,GSTs,AChE,CarE and AKP,were 1.68,1.68,1.27 and 1.62 times higher than those of the control,respectively.The results indicate that the homeostasis of C.suppressalis larvae was disrupted by the treatment of M.anisopliae CQMa421 preparation,which in turn exerted a toxic effect on the larvae.
