猕猴桃突变体库的构建及表型变异分析

为促进猕猴桃功能基因组学的研究,以'红阳'猕猴桃愈伤组织为试验材料,用 10、25、50、75、100、150、200、300、400、500 Gy共 10个不同辐射剂量的 60Co-γ射线处理愈伤组织,以 0 Gy为对照,对诱变后代的表型变异特征进行分析.结果表明:经100 Gy剂量诱变处理后,'红阳'猕猴桃愈伤组织的致变率已提升至35.9%,同时分化率亦达到 77.8%;基于此,筛选出 100 Gy为该品种愈伤组织的适当诱变条件;用 100 Gy诱变条件处理1 680块'红阳'猕猴桃愈伤组织,构建了由 1 026株再生苗组成的'红阳'猕猴桃突变体库,该突变体库在叶片、株型等性状上均存在丰富的变异,总变异频率达 35.9%.
Construction of a kiwifruit mutant library and analysis of phenotypic variations
To accelerate the development of functional genomic research,the mutant library of'Hongyang'kiwifruit was constructed.The callus of'Hongyang'kiwifruit were treated with 60Co-γ rays at 10 distinct radiation doses:10,25,50,75,100,150,200,300,400,and 500 Gy,with 0 Gy serving as the control,and the phenotypic variation characteristics of the mutagenic progeny were analyzed.The results indicated that after mutagenic treatment with a dose of 100 Gy,the mutation rate of callus tissue from'Hongyang'kiwifruit increased to 35.9%,while the differentiation rate reached 77.8%.Based on these findings,a radiation dose of 100 Gy was selected as the further studied condition.After treating 1 680'Hongyang'kiwifruit callus under optimal mutagenesis conditions,a kiwifruit mutant library contained 1 026 regenerated seedlings of'Hongyang'kiwifruit was constructed.The mutant library had rich variation in leaf,plant type and other characters,with a total variation frequency of 35.9%.
