Optimization of the Method for Determination of NDF and ADF in Tobacco with Detergent Method
In order to reduce the dosage of high toxic reagent of benzene,shorten the time of the degreasing process, and reduce the interference of incomplete hydrolysis of hemicellulose in washing process with acid detergent method, ethanol-ether mixture solvent was used for sample pretreatment after ultrasonic degreasing,and 2 mol/L HCl was used to wash the sample, then the acid detergent fiber was determined. Compared with general method,the optimization method had advantages of reducing the reagent toxicity, shortening the time from 8.5 h to 1.5 h, using 2 mol/L HCl to determinate the acid detergent fiber,which make hydrolysis more complete. Detergent method was used to calculate the hemicellulose content,the results were consistent with those of hydrolysis method. The recoveries were 98.38 %-99.97%,and the relative standard deviations of detection results were 0.25%-1.88%(n=6). The method is suitable for the determination of neutral detergent fiber and acid detergent fiber in tobacco and tobacco products instead of the mixture-solvent (containing benene, ethanol and ether) heating reflux method.
