Effects of total saponins of Panax notoginseng on re-differentiation of Treg into Th17 cells
OBJECTIVE To investigate the effect of total saponins of Panax notoginseng(PNS)on the trans-differentiation of Treg into Th17.METHODS The spleen cells of C57/BL6 mice were isolated and CD4+T cells were cultured in vitro by magnetic beads sorting.The appropriate concentration of PNS was screened by CCK-8 method,based on the index of cell viability.The cell model of Treg trans-differentiation into Th17 was established.The effect of PNS on the differentiation ratio of Treg/Th17 cells was detected by using flow cytometry,the level of Th17 cell characteristic cytokine IL-17A was measured by ELISA,and the Th17 cell characteristic transcription factor RORγt mRNA and inflammatory factor IL-17 mRNA was detected by real-time fluorescence quantitative polymerase chain reaction.RESULTS PNS could significantly inhibit the proportion of CD4+IL-17+Th17cells,the secretion of cytokine IL-17A and RORγt mRNA expression.CONCLUSION Certain concentrations of PNS can inhibit the trans-differentiation of Treg into Th17 and regulate the proportion balance of Treg/Th17 cells,and then prevent the occurrence and development of inflammatory immune diseases including rheumatoid arthritis.
