Study on the effects and mechanism of botanical compound MYD-20 against Staphylococcus aureus
OBJECTIVE To investigate the inhibitory effect and preliminary mechanism of the botanical compound MYD-20 and its ointment preparation on Staphylococcus aureus.METHODS The minimum inhibitory concentration(MIC)and minimum bactericidal concentration(MBC)of the clinical strains of Staphylococcus aureus and Methicillin-resistant S.aureus(MRSA)were determined by broth dilution method.The mechanism of antibacterial effect of MYD-20 was determined by the alkaline phosphatase activity and extracellular macromolecule concentration of the drug-treated bacterial solution as well as scanning electron microscopy.RESULTS MYD-20 showed strong antibacterial activity against S.aureus and MRSA,with MIC and MBC of 1.0%and 2.0%against both S.aureus and MRSA,respectively.AKP activity and extracellular macromolecule content in the bacterial solution were significantly increased after MYD-20 treatment,and combined with SEM observation,it could be found that the surface MYD-20 ointment showed significant wound healing effect and reduced the levels of inflammatory factors at the wounds in the model mice with deep Ⅱ degree skin scald infection.CONCLUSION The compound MYD-20 has a strong antibacterial effect on S.aureus and MRSA,and its mechanism is related to disrupting the permeability and integrity of bacterial cell membranes and cell walls,leading to leakage of cell contents.In addition,MYD-20 ointment can effectively reduce the level of inflammation at the wounds of scald-infected mice,as well as promote the healing effect of the wounds.
