Detection and optimization of ultrasonic extraction technology of gallic acid in Saorilao Qingfeizhike capsule
OBJECTIVE To detect and optimize the ultrasonic extraction technology of gallic acid in Saorilao Qingfeizhike capsule(SQC).METHODS The separation was conducted on a Hypersil BDS C18 column and the detection wavelength was 260 nm.The mobile phase consisted of methanol,acetonitrile and 0.1%phosphoric acid(5∶5∶90).Based on the gallic acid content,using methanol solution as the solvent,the four influencing factors of methanol concentration,ultrasonic temperature,material-liquid ratio and ultrasonic time were determined by single factor test and orthogonal test to determine the optimal extraction conditions.RESULTS The optimum extraction process was as follows:methanol concentration of 70%,ultrasonic temperature at 50 ℃,material-liquid ratio of 1∶80,ultrasonic time of 10 min.Gallic acid had a good linear relationship with the peak area in the range of 4.04-40.4 µg·mL-1(r=0.9999).The average content of gallic acid in SQC was 4.83 mg·g-1 by HPLC.CONCLUSION The optimized ultrasonic extraction conditions have proven to be simple and feasible with high gallic acid extraction and accuracy.
