乙醇-水为流动相的HPLC法测定氢氯噻嗪片的含量

摘要
目的 通过升高柱温降低柱压,建立较大流速的以乙醇-水为洗脱系统的HPLC法来测定氢氯噻嗪片的含量.方法 以0.1 mol·L-1磷酸二氢钠(用磷酸调pH3)-乙醇(88∶12)为流动相,采用Welch Ultimate C18色谱柱(250 mm × 4.6 mm,5 μm),柱温40℃,流速1.5 mL·min-1,检测波长271 nm.结果 0.0527~1.3185μg氢氯噻嗪与峰面积呈良好的线性关系,平均回收率为98.87%.结论 所用方法环保、准确,能替代氢氯噻嗪测定的现行HPLC法.
Abstract
OBJECTIVE To determine the content of Hydrochlorothiazide tablets by HPLC method with ethanol-water as the elution system.METHODS 0.1 mol·L-1 sodium dihydrogen phosphate(pH3.0)-ethanol(88∶12)was used as mobile phase.Welch Ultimate C18 column was used as the chromatographic column.The column temperature was 40℃;the flow rate was 1.5 mL·min-1and the detection wavelength was 271 nm.RESULTS Hydrochlorothiazide injection volume had a good linear relationship with peak area in the range of 0.0527-1.3185 μg,and the average recovery was 98.87%.CONCIUSION The method is environmentally friendly,accurate and can replace the current HPLC method for the determination of Hydrochlorothiazide tablets.
