LC-MS/MS法测定重镇过敏煎与过敏煎中的差异代谢物

目的 分析重镇过敏煎(过敏煎+磁石+龙骨)与过敏煎的差异代谢物,研究重镇过敏煎缓解特应性皮炎瘙痒的药效物质基础.方法 采用非靶向LC-MS代谢组学技术检测重镇过敏煎和过敏煎血清中的化学成分,基于多元统计分析筛选血清中的差异代谢物并进行通路分析.结果 正负离子两种模式下,两组血清中共检测出684种代谢物,筛选得到差异代谢物165种,主要为黄酮类、萜类、苯丙素类;富集得到2条具有生物学意义的通路,分别为苯丙氨酸代谢通路与苯丙氨酸、酪氨酸和色氨酸的生物合成通路.结论 重镇过敏煎对特应性皮炎瘙痒的治疗优势可能与苯丙素类、萜类、黄酮类等种类代谢物,以及azukisaponin Ⅳ、formononetin、6-shogaol、α-hederin等显著差异代谢物相关,以上代谢物通过参与调控苯丙氨酸代谢通路与苯丙氨酸、酪氨酸和色氨酸的生物合成通路实现.
Determination of metabolite differences between Zhongzhen Guomin decoction and Guomin decoction by LC-MS/MS
OBJECTIVE To study the pharmacodynamic material basis of Zhongzhen Guomin decoction[ZGD,Guomin decoction(GD),+Longgu+Cishi)]on relieving itchiness in atopic dermatitis by analyzing the differential metabolites of ZGD and GD.METHODS The chemical composition of serums of ZGD and GD was analyzed using Non-targeted LC-MS metabolomics,and differential metabolites were screened and pathway analysed between the two serum groups based on Multivariate statistical analysis.RESULTS Under the two modes of positive and negative ions,a total of 684 metabolites were detected in the serums of ZGD and GD.Among 165 significantly different metabolites,the types were mainly concentrated in flavonoids,terpenoids,phenylpropanes.Two pathways of obvious biological significance were found:phenylalanine metabolism pathway and phenylalanine,tyrosine and tryptophan biosynthesis pathway.CONCLUSION The therapeutic advantages of ZGD on atopic dermatitis pruritus may be related to the types of metabolites including phenylpropanoids,terpenoids,and flavonoids,as well as significant differences in metabolites such as batatasin Ⅳ,formononetin,6-shogaol and alpha-hederin.This is accomplished by participating in the regulation of the phenylalanine metabolic pathway and the biosynthesis pathway of phenylalanine,tyrosine and tryptophan.
