Optimization of water extraction process and in vitro antioxidant activity of Polygonati rhizoma extracts by Response surface methodology
OBJECTIVE To optimize the water extraction process of Polygonati Rhizoma extracts(PRE)and to study its in vitro antioxidant activity.METHODS We utilized the combination of subjective and objective weighting combined with the response surface method and HPLC.Taking material-liquid ratio,extraction time,and extraction times as inspection factors was adopted.We also took 6-gingerol,total polysaccharides content and dry extract yield as the evaluation indicators.We also aimed at optimizing the water extraction process of PRE,and in vitro antioxidant activity evaluation of PRE under the best technology.RESULTS The results showed that the optimal water extraction process of PRE was 1∶8 with the ratio of solid to liquid,the extraction times were 3 times,and the extraction time was 1.5 h.The relative error was 3.494%,which was close to the predicted value.The antioxidant activity test showed that the removal rate of DPPH free radicals and hydroxyl radicals increased with the mass concentration of the aqueous extract.At 50 mg·mL-1,the removal rate of hydroxyl free radicals and DPPH free radicals in the aqueous extract was 89.49%±0.02%and 83.80%±0.01%,respectively.CONCLUSION The optimized water extraction process is stable,reasonable and feasible,and the water extraction solution has the good antioxidant activity,which can provide the basis for the further development of PRE ointment food homologous products.
