Simultaneous determination of six tryptamine alkaloids in Hippophae rhamnoides by HPLC
OBJECTIVE To simultaneous determine the of 6 tryptamine alkaloids in Hippophae rhamnoides L.by HPLC.METHODS The separation was performed on Xcharge-C18(250 mm × 4.6 mm,5 μm)column with mobile phase consisted of methanol-0.15%trifluoroacetic acid solution by gradient elution at the flow rate of 1.0 mL·min-1.The wavelength was set at 280 nm,column temperature was 30 ℃ and the injection volume was 10 μL.RESULTS The linear ranges of 6 tryptamine alkaloids were 6.28-251.40 μg·mL-1,5.71-228.60 μg·mL-1,1.14-457.10 μg·mL-1,0.80-32.00 μg·mL-1,0.31-12.60 μg·mL-1and 0.46-18.30 μg·mL-1(r2>0.9993),respectively.Four kinds of tryptamine alkaloids were detected in the test samples,which were 5-hydroxytryptamine,6-hydroxy-l-methyltetrahydro-β-carboline-1-carboxylic acid,shepherdine and L-tryptophan.The average recoveries were 93.63%,89.86%,91.58%and 90.91%,respectively.The RSDs were less than 3.00%.CONCLUSION The method is simple and reliable,with good repeatability and accurate results.
