平胃胶囊对肝郁脾虚型反流性食管炎大鼠食管黏膜炎症的影响

目的 研究平胃胶囊对反流性食管炎模型大鼠核因子κB(NF-κB)/p65相关因子表达的影响.方法 采用食管十二指肠侧侧吻合术+饥饱失常+夹尾刺激制备大鼠肝郁脾虚型反流性食管炎模型.将大鼠分为正常组、模型组、平胃胶囊组(低、中、高剂量)、枳术宽中胶囊组、莫沙必利组,每日2次,共4周.镜下及肉眼观察评价食管的组织病变,ELISA法、Western Blot、Rt-qPCR法检测相关因子的表达水平.结果 平胃胶囊可显著改善模型大鼠食管的黏膜炎症,降低血清中炎性因子白细胞介素-1β(IL-1β)、肿瘤坏死因子-α、IL-6及食管组织中NF-κB抑制蛋白激酶β(IKKβ)、p65、环氧化酶2蛋白的水平,升高NF-κB抑制蛋白α(IKBα)表达.结论 平胃胶囊通过抑制胃组织中IKKβ/IκBα/NF-κB通路,降低血清中炎症因子的水平,减轻慢性胃炎大鼠的胃黏膜损伤.
Effects of Pingwei capsules on esophageal mucosal inflammation in Ganyu Pixu reflux esophagitis rats
OBJECTIVE To study the effect of Pingwei capsule on the expression of nuclear factor κB(NF-κB)/p65-related factors in reflux esophagitis model rats.METHODS Lateral esophagoduodenal anastomosis+hunger and satiety disorders+pinch-tail stimulation were used for the preparation of Ganyu Pixu reflux esophagitis model.It was divided into normal group,model group,low,medium and high dose group of Pingwei capsules,Zhizhu Kuanzhong capsules group,and mosapride group,twice a day for 4 weeks.Microscopic and naked eye observation was used to evaluate the esophageal tissue lesions,and the expression levels of related factors were detected by ELISA,Western Blot and Rt-qPCR.RESULTS Pingwei capsules could significantly improve the inflammation of esophageal mucosa in RE rats,reduce serum inflammatory factors such as IL-1β,TNF-α and IL-6,and downregulate esophageal tissue IKKβ,p65,COX2 protein levels,elevated IκBα expression.CONCLUSION Pingwei capsules reduce gastric mucosal injury in rats with chronic gastritis by inhibiting IKKβ/IκBα/NF-κB pathway in gastric tissues and lowering serum inflammatory factor levels.
