Protective effects of water-soluble royal jelly protein on high glucose induced human umbilical vein endothelial cells
OBJECTIVE To study the protective effect and mechanism of water-soluble royal jelly protein(WSRJP)on high glucose induced human umbilical vein endothelial cells(HUVEC).METHODS CCK-8 method was used to detect the effect of WSRJP on the proliferation activity of HUVEC.Enzyme labeling method was used to detect the effect of WSRJP on the secretion of superoxide dismutase(SOD).Enzyme-linked immunosorbent assay was used to detect the effect of WSRJP on the secretion of inflammatory factors tumor necrosis factor α(TNF-α)and interleukin-4(IL-4).Flow cytometry was used to detect the effect of WSRJP on the secretion of reactive oxygen species.Hoechst-PI double staining method was used to detect the effect of WSRJP on apoptosis and cell necrosis.RESULTS After treatment with WSRJP,HUVEC damaged by high glucose showed significant improvement in cell morphology and increased proliferation activity,and the reactive oxygen species secretion was increased.The ability to clear reactive oxygen species was improved,and the secretion of TNF-α and IL-4 was decreased.Apoptosis is reduced,but there is no effect on cell necrosis rate.CONCLUSION WSRJP can resist high glucose induced HUVEC damage,and its mechanism may be related to improving cell antioxidant capacity and reducing inflammatory response.
