目的 采用UPLC指纹图谱、化学计量学分析及定量分析,比较不同企业陈皮配方颗粒中橙皮苷和柚皮芸香苷的含量,并结合网络药理学预测其潜在质量标志物.方法 采用UPLC法测得23批陈皮配方颗粒的指纹图谱,结合化学计量学分析得到差异性成分,并测定其含量.通过网络药理学构建"成分-靶点-通路图"预测潜在质量标志物.结果 23批陈皮配方颗粒指纹图谱的相似度为0.937~0.999,确定5个共有峰,化学计量学分析得出质量差异化合物为橙皮苷和柚皮芸香苷,其含量测定结果B、D企业较高;网络药理学筛选得到橙皮苷、柚皮芸香苷、橘皮素、川陈皮素等化合物的15个核心靶点(TP53、AKT1、SRC等),通过调节松弛素信号、肿瘤和表皮生长因子受体等信号通路来治疗肿瘤、糖尿病,结合质量标志物原则得出橙皮苷、柚皮芸香苷、橘皮素、川陈皮素为潜在质量标志物.结论 所用方法可明确反映不同企业陈皮配方颗粒中差异化合物的含量,并采用网络药理学预测质量标志物,为陈皮配方颗粒的整体质量控制研究奠定了基础.
Determination of hesperidin and naringin in Citrus reticulata formula granules and analysis of their quality markers
OBJECTIVE To compare the content of hesperidin and naringin in Citrus reticulata formula granules(CRFG)obtained from different enterprises by UPLC fingerprint analysis,stoichiometry analysis and quantitative analysis,and predicting potential quality biomarkers based on network pharmacology.METHODS The fingerprint profiles of 23 batches of CRFG were obtained by means of stoichiometry analysis to identify the differential compounds and determine their contents.The component-target-pathway network diagram was constructed by network pharmacology to predict its potential quality markers.RESULTS The similarity of fingerprint between 23 batches of CRFG ranges from 0.937 to 0.999,and five common peaks were identified,from which the naringin and hesperidin as differential components were selected by stoichiometry analysis.The content determination results showed that the content of enterprise B and D was higher.Network pharmacologic screening analysis identified that TP53,AKT1 and SRC and other 15 core targets of the four compounds regulated the signaling pathways of relaxin,cancer and epidermal growth factor receptor to cure diabetes and other diseases.Combined with the principle of quality markers,hesperidin,narirutin,tangeretin and naringin were identified as potential quality markers.CONCLUSION The method can clearly reflect the content of different compounds in CRFG from different enterprises,and predict quality markers by network pharmacology,which lays a foundation for the overall quality control of CRFG.
