目的 优化唐古特大黄抗炎成分组(RTAC)的纯化工艺及其抗炎作用研究.方法 以唐古特大黄中的10种抗炎活性成分作为抗炎活性成分组,包括番泻苷B、芒柄花苷、番泻苷A、大黄素-8-O-葡萄糖苷、芒柄花素、芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚等.采用Agilent TC-C18色谱柱(250 mm ×4.6 mm,5 μm),以0.1%甲酸溶液-甲醇为流动相,梯度洗脱,流速1 mL·min-1,柱温30℃,进样量10 μL,检测波长280 nm.用溶剂法和大孔吸附树脂提取RTAC并进行分离纯化,考察其抗炎活性.结果 RTAC中的10种活性成分主要存在于二氯甲烷和水饱和正丁醇部位;水饱和正丁醇部位用大孔吸附树脂进一步纯化后,RTAC的含量从6.27%升至40.08%.纯化后的RTAC各剂量均可显著降低小鼠耳、足、棉球肉芽的肿胀度,且高剂量组的效果显著优于纯化前的唐古特大黄甲醇提取物组.结论 所用方法可用于分离纯化唐古特大黄的抗炎活性成分,为药材的的开发利用提供了依据.
Optimization of the purification process for anti-inflammatory constituents of Rheum tanguticum and study on its anti-inflammatory effects
OBJECTIVE To optimize the purification process of anti-inflammatory constituents group from Rheum tanguticum(RTAC)and to study its anti-inflammatory effects.METHODS The 10 kinds including sennoside B,ononin,sennoside A,emodin 8-O-glucoside,mangostensin,aloe emodin,rhein,emodin,chrysophanol,and emodin methyl ether were used as RTAC.The HPLC analysis was performed on an Agilent TC-C18 chromatography column(250 mm ×4.6 mm,5 µm).The mobile phase consisted of 0.1%formic acid aqueous solution-methanol.The flow rate was 1 mL·min-1,and the column temperature was 30 ℃ with the injection volume of 10 μL and the detection wavelength of 280 nm.The extraction,separation and purification of RTAC were used by the solvent method and macroporous adsorption resin method,and the activity of RTAC was investigated.RESULTS 10 components in the RTAC of mainly existed in the dichloromethane and water saturated n-butanol parts.The water saturated n-butanol part was further purified with macroporous adsorption resin,and its mass fraction increased from 6.27%to 40.08%.The purified RTAC with each dose significantly reduced the swelling of mice ears,feet,and cotton ball granulation,and the high-dose group had significantly better effects than the pre-purified R.tanguticum methanol extracts group.CONCLUSION This method can be used to separate and purify the RTAC,providing a basis for the further development and utilization of Rheum tanguticum.
