5种蓟属药用植物的ITS2序列分析

目的 对5种蓟属药用植物58份样品的ITS2序列进行分子鉴定,探讨蓟属药用植物的亲缘关系.方法 采用植物基因组DNA提取试剂盒提取58份样本中的DNA,对ITS2序列进行PCR扩增并测序;用MEGA 7.0计算其种间、种内的K2P距离,构建其NJ系统发育树.结果 5种蓟属药用植物的ITS2序列长度为227~228 bp,GC含量为59.65%~63.60%;种内最大遗传距离为0.0089,种间遗传距离为0.0179~0.0796;ITS2序列的二级结构中主环、Ⅰ区和Ⅲ区茎环的数目和大小可有效区别;NJ系统发育树分析发现:莲座蓟和绒背蓟的亲缘关系较近.结论 ITS2序列可作为5种蓟属药材鉴别的DNA条形码序列.
ITS2 sequence analysis of 5 medicinal plants of Cirsium
OBJECTIVE To identify the ITS2 sequences of 58 samples from 5 species of medicinal plants of Cirsium by DNA barcode technology,and to discuss the genetic relationship of medicinal plants of Cirsium.METHODS The total DNA was extracted from 58 samples of 5 species of Cirsium using the plant genome DNA extraction kit,and the ITS2 sequence was amplified by PCR and sequenced.The interspecific and intraspecific Kimura 2-parameter distances were calculated with Mega 7.0 to construct its neighbor-joining phylogenetic tree.RESULTS The ITS2 sequence length of five species of Cirsium medicinal plants was 227-228 bp,and the GC%content ranged from 59.65%to 63.60%.The maximum intraspecific genetic distance was 0.0089,and the interspecific genetic distance was 0.0179-0.0796.Through the secondary structure of ITS2 sequence,the number and size of stem rings in region Ⅰ and region Ⅲ can be effectively distinguished.NJ phylogenetic tree analysis showed that the genetic relationship between Cirsium esculentum and Cirsium vlassovianum is close.CONCLUSION ITS2 sequence can be used as DNA barcode sequence for identification of five medicinal materials of Cirsium.
