大鼠血浆中米托坦的测定及其药动学研究

目的 测定大鼠血浆中米托坦的含量,并研究米托坦在大鼠体内的药动学.方法 用乙腈沉淀预处理血浆样品,采用HPLC法检测米托坦在大鼠血浆中的含量,运用DAS3.0软件分析药动学数据.结果 米托坦回归方程为:Y=1.296 ×105X+5.823 × 103(r2=0.9999),线性范围 0.025~8.008 μg·mL-1;绝对回收率、方法回收率分别为 97.64%~99.98%、99.93%~102.09%,精密度RSD均≤ 1.01%(n=3).米托坦静注与灌胃给药的Cmax有显著性差异.结论 所用方法符合生物样品分析的基本要求,适合用于测定米托坦在大鼠体内的含量.采用静注给药可显著提高米托坦的生物体内达峰浓度.
Determination and pharmacokinetic study of Mitotane in rats plasma
OBJECTIVE To determinate the Mitotane in rats plasma and to study perform pharmacokinetics of Mitotane in rats.METHODS The rats plasma samples underwent pretreatment using the organic solvent precipitation method with acetonitrile as the precipitation agent.The concentration of Mitotane was determined using HPLC,and the resulting pharmacokinetic data were analyzed using DAS 3.0 software.RESULTS The regression equation of Mitotane was Y=1.296× 105 X+5.823×103(r2=0.9999),with the linear range of 0.025-8.008 µg·mL-1.The absolute and method recoveries were between 97.64%and 99.98%,and between 99.93%and 102.09%,respectively.The precision RSD was ≤1.01%(n=3).The Cmax of Mitotane iv differed significantly from that of ig.CONCLUSION The HPLC analysis of Mitotane plasma concentration meets the basic requirements for biological samples,and is suitable for the determination of mitotane in rats.Intravenous administration can significantly increase the peak concentration of Mitotane in the organ.
