UPLC-MS/MS鉴定北葶苈子的化学成分

目的 采用超高效液相色谱-质谱联用法快速鉴定北葶苈子药材的化学成分.方法 采用Waters UPLC HSS T3色谱柱(100 mm ×2.1 mm,1.8 µm),柱温40℃,以甲醇-0.1%甲酸为流动相,梯度洗脱,在正负离子模式下采集高分辨质谱数据.结果 通过保留时间、高分辨精确分子量、MS/MS碎片等信息,结合对照品数据及文献对比分析,共鉴定了 95个化合物,包括黄酮类24个、脂肪酸类19个、苯丙素类14个、硫苷及异硫氰酸类11个、尿苷5个、其他类22个.结论 快速地分析北葶苈子中化学成分,总结了化合物的裂解规律,为其成分分析、质量控制、临床应用等提供参考.
Analysis of chemical components of Lepidium apetalum by UPLC-MS/MS
OBJECTIVE To rapidly identify the chemical constituents in Lepidium apetalum Willd.by UPLC-Q-Exactive Orbitrap MS.METHODS Waters UPLC HSS T3 column(100 mm × 2.1 mm,1.8 μm)was used,with column temperature of 40 ℃.Gradient elution was performed with methanol-0.1%formic acid as the mobile phase,and the high-resolution mass spectrometry data were collected in positive and negative ion modes.RESULTS Based on the retention time,high-resolution accurate molecular weight,MS/MS fragmentation and other information,combined with reference substance data,95 compounds were identified from L.apetalum,including 24 flavonoids,19 fatty acids,14 phenylpropanoids,11 glucosinolates and isothiocyanates,5 uridines,and 22 others.CONCLUSION The method can quickly and relatively resolved the chemical constituents in L.apetalum,summarize the cleavage law of the compounds.This study provides reference and scientific basis for the component analysis,quality control and clinical application of L.apetalum.
