UPLC多波长法测定补肾益脑胶囊中的10种成分

目的 采用超高效液相色谱法(UPLC)同时测定补肾益脑胶囊中的10种成分.方法 采用Acquity UPLC BEH-C18色谱柱(100mm×2.1 mm,1.7 μm),流动相为乙腈-0.1%磷酸水溶液,进行梯度洗脱,流速0.3 mL·min-1,柱温30 ℃,进样量1 μL,检测波长分别为203 nm(人参皂苷Rg1、人参皂苷Re、人参皂苷Rb1、哈巴俄苷)、250 nm(补骨脂素、异补骨脂素、五味子醇甲、五味子酯甲)、320 nm(阿魏酸、斯皮诺素).结果 阿魏酸、斯皮诺素、补骨脂素、异补骨脂素、人参皂苷Rg1、人参皂苷Re、哈巴俄苷、人参皂苷Rb1、五味子醇甲、五味子酯甲浓度分别在13.15~210.40、2.58~41.35、1.16~18.58、1.64~26.22、6.03~96.41、7.21~115.30、1.28~20.48、24.50~392.00、1.16~18.64、2.42~38.68 μg·mL-1时与峰面积呈良好的线性关系,其r均大于0.9998,平均回收率分别为92.8%、92.0%、97.3%、97.8%、91.0%、102.3%、95.3%、99.3%、101.4%、96.7%,RSD 分别为 1.30%、1.78%、2.65%、1.95%、1.86%、3.12%、1.09%、1.35%、1.60%、1.85%.结论 所用方法简便可靠、重复性好,可用于补肾益脑胶囊中10种成分的含量测定.
Determination of 10 components in Bushen Yinao capsules by UPLC with multi-wavelength
OBJECTIVE To determine the 10 components in Bushen Yinao capsules(BYC)by UPLC.METHODS The chromatographic column was Acquity UPLC BEH-C18(100 mm × 2.1 mm,1.7 μm).The mobile phase was acetonitrile-0.1%phosphoric acid aqueous solution.Gradient elution was performed at the flow rate of 0.3 mL·min-1,the column temperature was 30 ℃,and the sample volume was 1 μL.The detection wavelengths were 203 nm,250 nm,and 320 nm,respectively.RESULTS The mass concentrations of ferulic acid,spinosin,psoralatin,isopsoralatin,ginsenoside Rg1,ginsenoside Re,harbacoside,ginsenoside Rb1,schisandrin and schisantherin A were 13.15-210.40,2.58-41.35,1.16-18.58,1.64-26.22,6.03-96.41,7.21-115.30,1.28-20.48,24.50-392.00,1.16-18.64,1.16-18.64,2.42-38.68 μg·mL-1.There was a good linear relationship with the peak area,and the r were greater than 0.9998.The average recoveries were 92.8%,92.0%,97.3%,97.8%,91.0%,102.3%,95.3%,99.3%,101.4%,96.7%,and the RSD values were 1.30%,1.78%,2.65%,1.95%,1.86%,3.12%,1.09%,1.35%,1.60%,1.85%,respectively.CONCLUSION The method is simple,reliable and reproducible,and can be used for the determination of 10 components in BYC.
