fAFLP analysis on population genetic diversity of Eleutheronema rhadinum along China coast
Eleutheronema rhadinum is a warm marine fish,mainly distributed in the east and south of China coast.E.rhadinum is a valuable marine species with wide adaptability to salinity,fast growth,delicate and delicious meat,and its meat is rich in inorganic salt minerals such as calcium,phosphorus,iron,iodine and vitamins.At present,reports about E.rhadinum are concentrated in the aspects of taxonomy,resource investigation,physiology and morphology.Some research has been done on genetic diversity of E.rhadinum populations,but the sampling area is relatively scattered and not representative.In this paper,samples of 214 individuals from eight geographical populations of E.rhadinum were collected and amplified to analyze the genetic construction by fluorescent amplified fragment length polymorphism(fAFLP)labeling technology.With nine primer combinations,a total of 493 bands were generated,including 443 polymorphic,and the percentage of polymorphic bands was 89.86%.The mean values of the observed number of alleles(Na),effective number of alleles(Ne),Nei genetic diversity index(H),and Shannon information index(I)of the eight populations were 1.679 0,1.467 2,0.250 8 and 0.363 1.The order of geographical population genetic diversity was Zhoushan>Chongming>Qidong>Dongshan>Guangzhou>Zhanjiang>Qionghai>Gaoxiong.The AMOVA was used to analyze the genetic variation sources of eight populations,and the total genetic differentiation index(Fst)was 0.237 2,indicating that 23.72%of genetic variation came from inter-population.The genetic distance(D)between eight geographical populations ranged from 0.021 6 to 0.194 4 and the similarity coefficient(H)was between 0.823 4 and 0.978 6.Based on research data,the eight populations of E.rhadinum could be divided into four branches by the UPGMA tree.The Chongming population,Qidong population and Zhoushan population were clustered into one branch,Dongshan population and Guangzhou population were clustered into one branch,Gaoxiong group was clustered into one branch,Zhanjiang group and Qionghai group were clustered into one branch.The genetic diversity of E.rhadinum in the coastal waters of China was analyzed systematically by using fAFLP molecular markers,in order to provide basic research data for the assessment and conservation of wild resources of E.rhadinum,and its genetic breeding and artificial breeding.
