Objective To investigate the effect of puerarin on ulcerative colitis(UC)induced by dextran sodium sulfate(DSS)in mice and its mechanisun.Methods 2.5%DSS solution was administered to mice to construct UC model.40 mice were randomly divided into normal group,model group,low dose group(20mg/kg·d),medium dose group(30mg/kg·d)and high dose group(40mg/kg·d).After administration for 8 days,the changes of body weight,disease index(DAI)and colon length were evaluated.The degree of colon tissue injury was evaluated by hematoxylin-eosin staining,and the levels of interleukin-6(IL-6),interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α),myeloperoxidase(MPO),nitric oxide(NO)and inducible nitric oxide synthase(iNOS)in colon tissues of mice were quantitatively detected by enzyme-linked immunosorbent assay kit.The level of synthase(iNOS).Results Puerarin could slow down the trend of weight loss,decrease the disease activity index,delay the shortening of colon length and reduce the degree of colon tissue injury.Preliminary study on the mechanism showed that different puerarin dose groups could reduce the levels of IL-6,IL-1β,IL-α,MPO,NO and iNOS.Conclusion Puerarin has protective effect on DSS induced UC mice,and its mechanism may be related to anti-inflammation and anti-oxidation.
