Objective To study the improvement effect of paeonol on oxidative stress in the rat oocytes in vitro maturation system,to determin the optimal concentration of paeonol for improving oxidative stress,to optimize the in vitro culture system and improve production efficiency.Methods 2mmol/L,4mmol/L,6mmol/L,8mmol/L and 10mmol/L of paeonol were added to the vitro maturation system to study the maturation quality and oxidative stress level of the rat oocytes in vitro maturation system.Results The addition of paeonol could significantly improve the maturation rate of oocytes compared to the control group(P<0.05).The GVBD rate,fertilization rate and blastocyst rate of oocytes in the 8 mmol/L paeonol group were higher than those of other experimental groups(P<0.05).The levels of reactive oxygen species in the 6 mmol/L,8 mmol/L and 10 mmol/L experimental groups were relatively low,and there were significant difference compared to the 2 mmol/L and 4 mmol/L experimental groups(P<0.01).Adding paeonol can significantly increase the expression levels of BCL-2 gene and protein in mature oocytes,and reduce the expression levels of BAX gene,Caspase-3 gene and protein(P<0.01).The groups of 8 mmol/L and 10 mmol/L showed significant differences compared to other groups(P<0.01).Conclusion Adding 8 mmol/L of paeonol to the in vitro maturation solution of rat oocytes could improve the production efficiency and quality of in vitro maturation of oocytes.It could also effectively improve the oxidative stress levels,and reduce the apoptosis of rat oocyte.
