为探究外源褪黑素(Melatonin,MT)在肉牛体内代谢情况及其对2次氯前列醇钠法处理母牛同期发情率、情期受胎率、血液抗氧化水平和生殖激素的影响,本研究首先将12头牛随机分为4组,早8时注射0 mg、0。464 mg、4。64 mg和46。4 mg褪黑素,测定不同时间点血液褪黑素浓度;其次将80头牛随机分为4组,在第2次氯前列醇钠注射和人工输精时,注射0 mg、0。464 mg、4。64 mg和46。4 mg褪黑素,统计发情率和情期受胎率,测定血浆抗氧化水平和生殖激素含量。结果表明,肉牛皮下注射不同剂量褪黑素后,血浆褪黑素在0。5 h处于较高值且极显著高于对照组(P<0。01),然后呈下降趋势,其中注射0。464 mg褪黑素经8 h代谢至正常水平,4。64 mg或46。4 mg褪黑素经14 h代谢至正常水平;同期发情处理中,皮下注射46。4 mg褪黑素可以显著提高肉牛发情率(P<0。05),情期受胎率呈现高于对照组的趋势;4。64 mg或46。4 mg褪黑素处理可显著降低血浆丙二醛(MDA)的含量(P<0。05),显著提高谷胱甘肽过氧化物酶(GSH-Px)的含量(P<0。05),极显著提高超氧化物歧化酶(SOD)的含量(P<0。01);46。4 mg褪黑素处理可显著提高配种时血浆中促卵泡素(FSH)和促黄体生成素(LH)的浓度(P<0。05),4。64 mg褪黑素可以显著提高配种时FSH和雌二醇(E2)的浓度(P<0。05),并且显著提升配种后第7天血浆孕酮(P4)值(P<0。05)。本研究表明,外源褪黑素通过提升机体抗氧化水平和改变生殖激素浓度提高2次氯前列醇钠法母牛同期发情率和情期受胎率,以注射46。4 mg褪黑素的效果最好。
Effects of Melatonin on Estrus Synchronization and Conception Rate in Beef Cattle
The aim of this study was to investigate the metabolism of melatonin(MT)in beef cattle and the effects of MT on the synchronous estrus rate,conception,blood antioxidant level and reproductive hormones.In this study,firstly,twelve beef cattle were randomly divided into four groups,0 mg,0.464 mg,4.64 mg and 46.4 mg melatonin were injected at 8 o'clock in the morning and the melatonin concentra-tions at different time pionts were measured.Then,eighty beef cattle were randomly divided into four groups,melatonin of 0 mg,0.464 mg,4.64 mg or 46.4 mg was injected during the second injection of clo-prostenol sodium and artificial insemination in the treatment,and estrus rate and estrus conception rate were counted.The blood was collected for the determination of plasma antioxidant level and reproductive hormone content.Results showed that after different doses of melatonin injected subcutaneously,plasma melatonin was at a high value and significantly higher than the control group at 0.5 h(P<0.01),then it begen to decline.0.464 mg melatonin injected subcutaneously into beef cattle was metabolized to the nor-mal level through 8 h,while 4.64 mg or 46.4 mg melatonin to the normal level through 14 h.When me-latonin was injected subcutaneously during estrus synchronization,46.4 mg melatonin injected subcutane-ously could significantly improve the estrus rate of beef cattle(P<0.05),and the conception was higher than that of the control group.4.64 mg or 46.4 mg melatonin could significantly reduce the content of MDA(P<0.05),increase the content of GSH-PX(P<0.05),and significantly increase the content of SOD(P<0.01).46.4 mg melatonin could significantly increase the concentration of FSH and LH in plas-ma during mating(P<0.05).4.64 mg melatonin could significantly increase the concentration of FSH and E2 during mating(P<0.05),and can increase the P4 content on the 7th day after insemination(P<0.05).This study shows that exogenous melatonin can enhance the synchronous estrus rate and conception in two cloprostenol sodium treatments by improving antioxidant level and reproductive hormone concentration of beef cattle,and the effect is best when 46.4mg melatonin is injected subcutaneously into beef cattle.
