14个山羊群体FGF5座位c.-253G>A和507 bp缺失的遗传分析

摘要
为鉴定与绒山羊绒毛长度性状有关的分子标记,为遗传改良提供理论依据,该研究利用PCR-RFLP对来自14个群体的608只山羊FGF5座位c.-253G>A和507 bp缺失进行基因分型,检测2个位点的等位基因频率,并分析其与海拔、年平均气温的相关性.结果表明,在5个绒山羊群体和3个肉绒兼用藏山羊群体中,c.-253G>A位点的突变A等位基因和507 bp缺失位点的Mut等位基因频率分别为60.9%~98.3%和95.7%~100%.在6个低海拔肉皮兼用或肉用群体中,A和Mut等位基因的频率分别为0~51.3%和0.7%~47.5%.除昌都藏山羊群体,13个山羊群体在2个位点均处于Hardy-Weinberg平衡.2个突变位点等位基因频率与海拔高度和年平均气温均呈极显著相关(P<0.001).由此可知,FGF5座位c.-253G>A和507 bp 2个突变位点等位基因频率在高海拔且气温较低地区的绒山羊品种中极高,应是调控山羊绒毛长度的重要位点,可作为绒山羊遗传改良的分子标记.
Abstract
To identify the genetic markers related to cashmere traits and provide theoretical references for marker-assisted selection in goats,in this study,608 goats from 14 populations were genotyped using PCR-RFLP technology to detect allele frequencies at the c.-253G>A and 507 bp deletion of the FGF5 lo-cus and analyze the correlation between the allele frequency and altitude and annual average temperature.Results showed the frequencies of the A allele at the c.-253G>A and the Mut allele at the 507 bp deletion sites were 60.9%~98.3%and 95.7%~100%in five cashmere goat populations and three populations of Tibetan goats with dual-purpose for meat and cashmere,respectively.In six low-altitude populations,the frequencies of the A and Mut alleles varied from 0 to 51.3%and from 0.7%to 47.5%.The results also showed that excepting for the Qamdo Black Goat population,all 13 goat populations were in Hardy-Wein-berg equilibrium at both loci.The mutant allele frequency of the two loci was significantly correlated with altitude and annual average temperature(P<0.001).These showed that the mutant allele frequencies at both c.-253G>A and 507 bp deletion of FGF5 locus were extremely high in cashmere goats living in the areas with high altitude and relatively low temperatures.Therefore,both loci are the important sites af-fecting the cashmere length and can be used as molecular markers for genetic improvements of cashmere goats.
