Goat semen is susceptible to damage from oxidative stress during storage.Quercetin,a fla-vonol compound widely found in plants,has several biological activities and has been shown to improve sperm quality.The aim of this experiment was to investigate the effect of quercetin addition to semen dilu-tions on semen quality of goats stored at low temperature(4℃).In this experiment,semen was collected from 1 to 3-year-old healthy Shaanxi Guanzhong dairy goats(n=9),and the antioxidant property,ATP content and related phosphorylated proteins of cryopreserved goat semen were analyzed after the addition of quercetin at different concentrations(10 μmol/L,25 μmol/L,50 μmol/L and 100 μmol/L).The results showed that the addition of quercetin could improve the viability of cryopreserved spermatozoa,protect the integrity of the plasma membrane and acrosome,inhibit the dephosphorylation of sperm proteins,and in-crease the expression level of phosphorylated proteins;the addition of quercetin could inhibit the dephos-phorylation of spermatozoa proteins induced by excessive reactive oxygen species(ROS)in semen through the cAMP-PKA signaling pathway.The optimal concentration of quercetin is 50 μmol/L.Namely,the ad-dition of 50 μmol/L quercetin to goat semen during cryopreservation can improve semen quality.
