摘要

本试验旨在分析长毛兔PRKACB基因序列遗传多态性及其与生产性状(体重、产毛量)的相关性,发现与产毛量相关的分子标记,为加快毛兔选育提供参考.试验采用直接测序法检测了 169 只皖系长毛兔PRKACB基因外显子区与启动子区序列的SNPs位点,进行遗传多样性分析,并运用SPSS 25.0 软件进行 SNPs 与体重、产毛量的关联性分析.结果显示,长毛兔PRKACB共有 10 个外显子和 9 个内含子,其中外显子序列较为保守,未检测到SNPs.启动子序列(1366bp)存在 2 个 SNP 位点(rs81065607A>C和rs81066149C>T),均包含 3 种基因型,均符合哈迪-温伯格平衡.rs81065607A>C和rs81066149C>T的PIC分别为 0.331 和 0.327,均为中度多态,提示PRKACB基因在群体中具有较高遗传变异能力.SNP 位点间连锁不平衡程度结果显示,2 个位点高度连锁(r2=0.983).关联分析表明,rs81065607A>C 的CC型个体的产毛量显著高于AA型和AC型(P<0.05);rs81066149C>T的TT型个体的产毛量显著高于CC型和CT型(P<0.05).结果表明,PRKACB 基因 rs81065607A>C和 rs81066149C>T位点可以作为长毛兔产毛量的潜在分子标记,用于长毛兔的分子选育.

Abstract

The objective of this study was to analyze the genetic polymorphism of PRKACB gene se-quence in Angora rabbits and its correlation with production traits(body weight,wool yield),and to iden-tify molecular markers in relation to wool production,and to provide reference for accelerating the breed-ing of Angora rabbits.Direct sequencing was used to detect SNPs in the PRKACB gene exon and promoter region of 169 Angora rabbits,and SPSS 25.0 software was used to analyze the correlation between SNPs and weight and wool production.Results showed PRKACB gene has 10 exons and 9 introns,with the exon sequences being relatively conserved and no SNPs detected.In the 1366bp promoter sequence,two SNP lo-ci(rs81065607 A>C and rs81066149 C>T)were found,both containing three genotypes and conforming to Hard-Weinberg equilibrium.The polymorphic information content values of rs81065607 A>C and rs81066149 C>T were 0.331 and 0.327,indicating moderate polymorphism,suggesting that the PRKACB gene has high genetic variability in the Angora rabbit population.The linkage disequilibrium analysis showed a high linkage between the two SNP loci(r2=0.983).The association analysis showed that the in-dividuals with the CC genotypes of rs81065607 A>C had significantly higher wool production than those with AA and AC genotypes(P<0.05);individuals with the TT genotype of rs81066149 C>T had signifi-cantly higher wool yield than those with CC and CT genotypes(P<0.05).It is therefore concluded that the SNP sites rs81065607 A>C and rs81066149 C>T in the PRKACB gene can be potential molecular markers for wool yield in Angora rabbits,and can be used in molecular breeding of Angora rabbits.
