To investigate the protective effect of Danqijing Granule on pulmonary vascular endothelium under acute high altitude exposure
Objective To explore the protective effect of Danqijing granules on pulmonary vascular endothelium after acute exposure to high altitude by network pharmacological methods and clinical trials. Methods The active ingredients and drug targets of Danqijing granules were collected by TCMSP platform, and the targets of disease action were retrieved to obtain the targets of disease-drug interaction.GO enrichment and KEGG pathway analysis of disease and drug crossover targets were then performed. Finally, the target network diagram of drug-disease pathway was constructed. The 80 patients were randomly divided into Rhodiola capsule control group(40 cases)and Danqijing granule group(40 cases). The levels of VEGF and ET-1 were recorded on the 1st,3rd and 7th day after the two groups were stationed at the plateau. Results The network pharmacological analysis showed that Danqijing granules may participate in the biological processes such as inflammation by regulating the VEGF signaling pathway in acute altitude exposure. The clinical trial results showed that compared with Rhodiola Rosea capsule group, the content of VEGF in Danqijing granule group was significantly increased(F=8.977,P<0.05),and the content of ET-1 was significantly decreased(F=5.248,P<0.05).Conclusion Danqijing Granule has a multi-target and multi-pathway therapeutic effect on acute high-altitude exposure, and can improve the dynamic levels of VEGF and ET-1,the core targets of vascular endothelial function, and protect pulmonary vascular endothelial cells.
