Difference in Effects of Hypoxia on Gene Expressions of Six Drug Transporters in Rats
Objective To study the gene expressions of six drug transporters-Pept1,Oatp1b1,Oat1,Oct1,Mdr1 and Mrp2 in rats after acute hypoxia at high altitude.Methods Wistar rats were randomly divided into normal control group,hypoxia 24 h group and hypoxia 72 h group.The experimental groups (hypoxia 24 h and 72 h groups) were fed in a hypobaric chamber simulated to an altitude of 5000 m (pressure:397.5 mmHg,the oxygen partial pressure:42 mmHg).Total RNA was extracted from the small intestine,liver and kidney of rats in each group.Real-time PCR was used to detect the expression levels of six transporter genes.Results Compared with control group,the expressions of most of the drug transporters Pept1,Oatp1b1,Oat1,Oct1,Mdr1 and Mrp2 in hypoxia groups were increased,but different transporters in different tissues developed diverse trends with the extension of hypoxia time.Conclusion Hypoxia at high altitude can affect the mRNA expression levels of drug transporters,which may provide some reference for the study of drug transporters and pharmacokinetics at high altitude.
