Roles of Suanzaoren Decoction in Improving Anxiety-like Behavior Induced by M-chloropiperazine in Mice
Objective To compare the effect of different formulations of Suanzaoren decoction on anxie-ty-like behavior of mice induced by single intraperitoneal injection of m-chlorpiperazine.Methods After seven days of administration of different formulations of Suanzaoren decoction,the extent to which Suanzaoren de-coction improved the anxiety-like behavior induced by m-chlorpiperazine was detected via the elevated-plus maze test.The contents of serum 5-hydroxytryptamine、corticosterone、adrenocorticotropic hormone and 5-hydroxytryptamine in the brain were detected by ELISA.Results In the elevated-plus maze test,single intraperitoneal injection of m-chlorpiperazine 4 mg·kg-1 induced anxiety-like behavior in mice.Suanzaoren decoction 21.7 g·kg-1 could prolong the time spent in open arms,increase the percentage of the time spent in open arms,and upregulate the content of 5-hydroxytryptamine in serum but downregulate the content of 5-hydroxytryptamine in the hypothalamus,amygdala,hippocampus and cortex.Serum adrenocorticotropic hormone and corticosterone were not affected by Suanzaoren decoction.Conclusion Suanzaoren decoction can improve anxiety-like behavior induced by single intraperitoneal injection of m-chlorpiperazine,and the mecha-nism may be related to regulating the serotonergic nervous system.
