摘要

目的 基于网络药理学和分子对接法探究罗布麻叶治疗高血压病的机制,为临床用药提供科学理论依据.方法 通过差异分析和加权基因共表达网络分析筛选高血压病相关基因集.依据中药药理学技术平台和SwissTargetPredic-tion数据库筛选罗布麻叶的潜在活性成分和预测潜在作用靶点.通过蛋白-蛋白网络分析和KEGG通路富集分析,探究罗布麻叶治疗高血压病的核心基因和关键信号通路.通过分子对接、实时荧光定量聚合酶链式反应和 Western blot验证筛选的核心基因.结果 通过网络药理分析和实验验证发现罗布麻叶治疗高血压病的关键基因SERPING1.基于KEGG通路富集分析发现,罗布麻叶治疗高血压病的关键信号通路是"补体和凝血级联反应"及"百日咳".结论 罗布麻叶可以调控SERPING1的表达通过"补体和凝血级联反应"及"百日咳"信号通路治疗高血压.

Abstract

Objective To investigate the mechanism by which Apocyni Veneti Folium combats hypertension based on network pharmacology and molecular docking in order to provide data for clinical medication.Methods Hypertension-related gene sets were screened via differential analysis and weighted gene co-expression network analysis(WGCNA).The active ingredients and potential Apocyni Veneti Foli-um action targets were identified based on TCMSP and SwissTargetPrediction databases.Subsequently,protein-protein network analysis and KEGG pathway enrichment analysis were performed to explore the core genes and critical signaling pathways of Apocyni Veneti Folium that were effective for hypertension.In addition,the screened core genes were validated via molecular docking,real-time quantitative PCR,and Western blot.Results Based on network pharmacological analysis and experimental validation,it was found that SERPING1 was the key gene involved in treating hypertension with Apocyni Veneti Folium.Moreover,Apocyni Veneti Folium could treat hypertensive diseases via'complement and coagulation cascades'and'Pertussis'pathways.Conclusion Apocyni Veneti Folium can be used to treat hypertensive diseases by upregulating SERPING1 and through'complement and coagulation cascades'and'Pertussis'pathways.
