摘要

目的 采用Box-Behnken响应面法优化加红生化方提取工艺.方法 在单因素试验基础上,以料液比、提取时间和提取次数为考察因素,以阿魏酸含量及配方浸膏得率为评价指标,Box-Behnken响应面法对提取工艺进行优化,并试验验证.结果 优化后提取工艺为料液比1∶8,提取时间1.5 h,提取次数为2次.在此条件下,测得的阿魏酸的含量为0.584 mg·g-1,浸膏得率为18.15%,综合评分为98.13(RSD=0.39%),与预测值98.92接近,偏差为0.80%.结论 该提取工艺稳定可靠,预测性良好,可为加红生化方相关中药制剂的研发提供参考.

Abstract

Objective To optimize the extraction process of Jiahong Shenghua prescription using the Box-Behnken response surface method.Methods Based on the single factor experiment,the solid-liquid ratio,extraction time and number of times of extractions were taken as factors for investigation,while the ferulic content and extract yield were chosen as the evaluation indices to optimize the extraction process with the Box-Behnken response surface method.Results The best extraction process involved a solid-liquid ratio of 1:8,extraction time of 1.5 h,and two-time extraction.Under this condition,the content of feru-lic and the extract yield were 0.584 mg·g-1 and 18.15%respectively.The comprehensive score was 98.13(RSD=0.39%),which was close to the predicted value of 98.92.The standard deviation was 0.80%.Conclusion This extraction process is stable,reliable and highly predictive,which can provide references for the development of traditional Chinese medicine related to Jiahong Shenghua prescription.
