摘要

目的 建立中药热敷包中羟基红花黄色素A的含量测定方法.方法 采用Agilent 1260高效液相色谱仪测定热敷包中羟基红花黄色素A的含量,样品用25%乙醇超声提取40 min.Kromasil C18色谱柱(4.6 mm× 250 mm,5 µm),柱温:30℃;流动相为乙腈-甲醇-0.7%磷酸水溶液(2∶26∶72),流速:1.0 ml·min-1;检测波长:403 nm.结果 羟基红花黄色素A在0.511~1.789μg范围内与其色谱峰面积有良好的线性关系(r=0.9999),该方法的平均回收率为98.65%,RSD为0.61%(n=6),3批中药热敷包中羟基红花黄色素A平均含量为0.83 mg·g-1.结论 本法稳定可靠,重复性好,可用于中药热敷包的质量检测.

Abstract

Objective To establish a quantitative determination method for a hot compressed package of traditional Chinese medicine.Methods The HPLC method was used to determine hydroxysafflor yellow A in the hot compressed package.Chromatographic conditions included Agilent 1260 HPLC,Kromasil C18(4.6 mm×250 mm,5 μm),a mobile phase composed of acetonitrile-methanol-0.7%phosphoric acid solu-tion(2∶26∶72),a velocity of 1.0 ml·min-1,a detection wavelength of 403 nm,and a column tempera-ture of 30 ℃.Results Within the range of 0.511-1.789 μg,the calibration cure was linear(r=0.9999).The mean recovery rate was 98.65%,with an RSD of 0.61%(n=6).The mean content of hydroxysafflor yellow A in the three batches of medicine was 0.83 mg·g-1.Conclusion This determination method is stable,accurate,reproducible,which can be used as a quality detection for hot compressed packages of traditional Chinese medicine.
