Mechanisms of Shouwu Xiaoban Capsules in the Treatment of Vitiligo Based on Network Pharmacology
Objective To investigate the mechanism of Shouwu Xiaoban capsules in the treatment of vitiligo by means of network pharmacology.Methods According to TCMSP database,HERB database,and SwissADME database,the main active ingredients were screened,and the SwissTargetPrediction data-base was used to predict the potential targets of the active ingredients.Vitiligo-related targets were obtained through the DisGeNET database and Gene Cards database.The Venny 2.1 online tool was used to obtain the intersections of the herbal medicine targets and the disease targets while Cytoscape 3.10.0 software and String database was employed to construct a drug-compound-target network and protein-protein interaction network.Finally,the data was analyzed visually by the online platform before DAVID database was used to annotate important targets by means of gene ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis.Results A total of 110 potential active ingredients of Shouwu Xiaoban capsules,834 related targets,1447 targets of vitiligo and 151 intersection targets were screened for the treatment of vitiligo with Shouwu Xiaoban capsules.Isoflavanone,baicalein,jaranol,pachypodol and 12-senecioyl-2E,8E,10E-atractylentriol were considered critical components while BCL2,AKT1,CASP3 and NFKB1 were recognized as key therapeutic targets.The GO analysis demonstrated that key the targets were mostly involved in the process of response to lipopolysaccharide,positive regulation of ERK1 and ERK2 cascade and protein phosphorylation.KEGG analysis suggested that the key targets were also related to pathways in cancer,EGFR tyrosine kinase inhibitor resistance,and C-type lectin receptors.Conclusion Shouwu Xiaoban capsules may exert therapeutic effects on vitiligo by regulating multiple components,targets,and pathways.This research is expected tospark new ideas for the treatment of vitiligo.
