The relationship between Kruppel-like transcription factor 8 and tamoxifen resistance in breast cancer
Objective To investigate the relationship of Krüppel-like factor 8(KLF8)on tamoxifen(TAM)resistance and prognosis of breast cancer.Method The expression difference of KLF8 mRNA in TAM sensitive vs drug resistant cells and the effect of KLF8 mRNA on prognosis of breast cancer patients treated with TAM were analyzed using public database.RT-PCR and Western blot were used to detect the expression differences of KLF8 mRNA and protein between MCF-7 and LCC2 cells.Immunohistochemistry was used to detect KLF8 expression in 97 patients was Luminal A/B subtype treated with TAM,and to analyze the relationship between KLF8 expression and prognosis.CCK-8 kit was used to detect the changes of cell proliferation under different concentrations of TAM after si-lencing or overexpressing KLF8.Results The expression level of KLF8 in TAM resistant cells was higher than that in TAM sensitive cells.In patients with Luminal A/B subtype breast cancer treated with TAM,patients with high KLF8 expression had poor prognosis,and the resistance to TAM was enhanced after overexpression of KLF8,while the resistance to TAM was weakened after silence of KLF8.Conclusion The high expression of KLF8 is associated with tamoxifen resistance in breast cancer.In Luminal A/B subtype of breast cancer,the high expression of KLF8 TAM has poor therapeutic effect.KLF8 may predict efficacy of tamoxifen and provide a new entry point for molecular targeted therapy.
