Changes of MicroRNAs expression profile after spinal cord injury in rats
Objective To explore the changes of microRNAs expression profile after spinal cord injury in rats.Method Ten adult SD rats were randomly divided into sham operation group and model group to establish the model of spinal cord injury.After operation,the motor function was evaluated by Basso-Beattie-Bresnahan(BBB)scale,and the spinal cord tissue was stained with hematoxylin-eo-sin(HE).The cDNA library was analyzed by RNA-Seq by Illumina high-throughput sequencing technique.After comparing the se-quencing data with the sorghum reference genome,the differentially expressed genes were screened under the condition of P<0.05.The function and regulatory pathway of DEGs were analyzed by GO,KEGG and other databases,and the reliability of RNA-Seq results was verified by RT-qPCR.Results There was significant difference in BBB score between sham operation group and spinal cord injury group.HE staining showed that the structure of spinal cord was seriously damaged,and there was significant difference in the expres-sion of microRNAs before and after spinal cord injury.Conclusion Many genes are dysregulated in spinal cord injury.
