Up-regulation of CX43 expression enhances the Bystander effect of suicide gene therapy for nasopharyngeal carcinoma
Objective To study the Bystander effect of upping CX43 expression to enhance suicide gene therapy for nasopharyngeal carcinoma.Method The recombinant plasmids carrying CX43 gene were constructed and transfected into nasopharyngeal carcinoma cells CNE-2 and CNE-2/TK,the difference of cell survival rate between CX43 non-transfected group and CX43 transfected group was compared,and the CNE-2/CNE-2 TK mixed cells(control group 1)was used as the control of CX43 non-transfected group,cNE-2-CX43/CNE-2 tk-Cx43 mixed cells(control group 2)were used as the control of CX43 transfected group.The difference of cell survival rate between CX43 non-transfected group and CX43 transfected group was compared.Results Control group 1 as control,the mixed ra-tios of tk-and tk+cells were 100∶0,95∶5,90∶10,80∶20,75∶25,50∶50,0∶100,respectively,the cell survival rates of CX43 non-trans-fected group and CX43 transfected group were 94.3%∶82.4%,83.2%∶63.5%,73.3%∶51.6%,52.7%∶33.9%,38.2%∶27.6%,17.6%∶16.8%,8.1%∶6.3%,respectively.When the mixture ratio of tk-and tk+cells was 100.0,95∶5,90∶10,80∶20,75∶25 respec-tively,there was significant difference between the two groups(P<0.05),while there was no difference in other groups(P>0.05)CNE-2/CNE-2 TK mixed cells(control group 1)were used as the control of CX43 non-transfected group,and CNE-2/CNE-2 TK mixed cells(control group 1)were used as the control of CX43 non-transfected group,when the mixture ratio of tk-and tk+cells was 100∶0,95∶5,90∶10,80∶20,75∶25,50∶50,0∶100,the cell survival rates of CX43 non-transfected group and CX43 transfected group were 91.8%∶94.7%,respectively,82.1%∶71.9%,72.6%∶58.1%,51.6%∶38.7%,35.5%∶29.8%,13.2%∶16.8%,7.6%∶6.3%.When the mixture ratio of tk-and tk+cells was 95∶5,90∶10,80∶20,75∶25 respectively,there was significant difference be-tween the two groups(P<0.05),while there was no difference in other groups(P>0.05).Conclusion The up-regulation of Cx43 and gap junction in NPC cells could obviously enhance the bystander killing effect of suicide gene therapy for NPC,and provide a new method for the prevention and treatment of NPC.
