Biological effect and molecular mechanism of ellagic acid on osteosarcoma MG-63
Objective To explore the biological effect and molecular mechanism of natural extract ellagic acid on the proliferation and apoptosis of osteosarcoma MG-63 cells,and to seek a new way to prevent and treat osteosarcoma.Method MG-63 cell lines were treated with 0 μg/ml,15 μg/ml,30 μg/ml,and 45 μg/ml of ellagic acid,Through Methyl thiazolyl tetrazolium assayand flow cytome-try,The effects of ellagic acid on proliferation,apoptosis and cell cycle of MG-63 cell line were analyzed.Western blotting was used to observe the changes of epidermal growth factor receptor(EGFR),cell proliferation and apoptosis factors Bax,Bcl-2 protein expres-sion.Results As the concentration of ellagic acid increased in the experimental group,the early apoptosis rate and inhibition rate of hu-man osteosarcoma MG-63 cells incubated with ellagic acid gradually increased in a concentration dependent manner;MG-63 cells trea-ted with tannic acid were arrested in G0/G1 phase;The expression of epithelialgrowth factor receptor(EGFR)decreased,the expres-sion of pro-tumor cell apoptosis factor Bax protein increased,and the expression of tumor apoptotic factor Bcl-2 protein decreased.Con-clusion Ellagic acid can inhibit the proliferation and induce apoptosis of MG-63 cell line,which may be related to the changes in the expression of epidermal growth factor receptor(EGFR),cytokines Bax and Bcl-2.
