β-防御素基因在乾华肉用美利奴羊生殖器官的先天免疫中发挥重要作用.为探究β-防御素基因对乾华肉用美利奴羊生殖系统的影响,分别提取雌性乾华肉用美利奴羊的卵巢、输卵管和子宫颈组织的总RNA,根据NCBI中绵羊β-防御素的cDNA序列设计并合成引物,反转录合成cDNA,实时荧光定量PCR检测β-防御素基因在雌性乾华肉用美利奴羊生殖系统中的表达水平.结果表明:β-防御素基因在雌性乾华肉用美利奴羊的卵巢、输卵管和子宫颈中均有表达,输卵管中的表达量最高,其次是卵巢,子宫颈中的表达量最低,各生殖器官中的表达量无显著差异(P>0.05).说明β-防御素基因在雌性乾华肉用美利奴羊生殖系统的先天免疫中发挥作用,为探究乾华肉用美利奴羊的抗病能力提供依据.
Expression Level of β-defensin in the Female Reproductive System of Qianhua Mutton Merino
β-defensin plays an important role in the innate immunity of reproductive organs of Qianhua mutton merino.In order to investigate the effect of β-defensin on the reproductive system of Qianhua mutton merino,total RNA was extracted from ovary,fallopian tube and cervix tissues of female Qianhua mutton merino.Primers were designed according to the cDNA sequence of β-defensin gene in NCBI,and the cDNA was synthesized by reverse transcription.Real-time fluorescence quantitative PCR was used to detect the expression level of β-defensin in the reproductive system of female Qianhua mutton merino.The results showed that β-defensin was expressed in ovaries,fallopian tubes and cervix of female Qianhua mutton merino,and the expression level in fallopian tube was the highest,followed by that in ovary,cervix was the lowest,and there was no significant difference in the expression level of reproductive organs(P>0.05).It is suggested that β-defensin plays a role in innate immunity of reproductive system of female Qianhua mutton merino,which provides basis for revealing the disease resistance of reproductive disorder of female Qianhua mutton merino.
