鸭绿江花羔红点鲑的DNA条形码及鉴定分析

应用DNA条形码技术对鸭绿江分布的花羔红点鲑样品进行鉴定.通过对鸭绿江花羔红点鲑的线粒体细胞色素氧化酶Ⅰ亚基(CO Ⅰ)基因序列进行分析,应用DNA条形码技术鉴定鸭绿江花羔红点鲑的种类.结果表明:花羔红点鲑线粒体CO Ⅰ基因序列长为699 bp,A,T,C,G碱基的含量分别为22.75%,29.61%,28.76%和18.88%,G含量较低,A+T含量(52.36%)高于C+G含量(47.64%).花羔红点鲑种内差异不明显,样品鱼种内遗传距离均<0.02.对样品鱼进行DN A基因序列分析,GenBank和Bold Systems搜索结果相似性在99.56%~100.00%,因此,分布于鸭绿江的红点鲑鉴定为花羔红点鲑(Salvelinus malma).
DNA Barcode and Identification Analysis of Salvelinus malma in Yalu River
The purpose of this study was to identify the species of Dolly Varden Trout in Yalu River by DNA barcoding technique.Through analyzing the gene sequence of cytochrome oxidase Ⅰ subunit(CO Ⅰ)of Yalu River Dolly Varden Trout,the species of Yalu River Dolly Varden Trout was identified by DNA barcoding.The results showed that the length of mitochondrial CO Ⅰ gene was 699 bp,and the contents of A,T,C and G bases were 22.75%,29.61%,28.76%and 18.88%,respectively,in which the content of G was lower,the content of A+T(52.36%)was higher than that of C+G(47.64%).There were no significant intraspecific differences and the average intraspecific genetic distance of the Dolly Varden Trout was less than 0.02.DNA gene sequence analysis showed that the highest similarity with the GenBank and Bold Systems was 99.56%-100.00%.A comprehensive analysis of intraspecific genetic distance and search results showed Dolly Varden Trout in the Yalu River was Salvelinus malma.
